    Decellularized scaffolds can serve as an excellent three-dimensional environment for cell repopulation. They maintain tissue-specific microarchitecture of extracellular matrix proteins with important spatial cues for cell adhesion, migration, growth, and differentiation. However, criteria for quality assessment of the three-dimensional structure of decellularized scaffolds are rather fragmented, usually study-specific, and mostly semi-quantitative. Thus, we aimed to develop a robust structural assessment system for decellularized porcine liver scaffolds. Five scaffolds of different quality were used to establish the new evaluation system. We combined conventional semi-quantitative scoring criteria with a quantitative scaffold evaluation based on automated image analysis. For the quantitation, we developed a specific open source software tool (ScaffAn) applying algorithms designed for texture analysis, segmentation, and skeletonization. ScaffAn calculates selected parameters characterizing structural features of porcine liver scaffolds such as the sinusoidal network. After evaluating individual scaffolds, the total scores predicted scaffold interaction with cells in terms of cell adhesion. Higher scores corresponded to higher numbers of cells attached to the scaffolds. Moreover, our analysis revealed that the conventional system could not identify fine differences between good quality scaffolds while the additional use of ScaffAn allowed discrimination. This led us to the conclusion that only using the combined score resulted in the best discrimination between different quality scaffolds. Overall, our newly defined evaluation system has the potential to select the liver scaffolds most suitable for recellularization, and can represent a step toward better success in liver tissue engineering.

Über das Gesicht des Menschen und über die Kunst, den menschlichen Charakter und alles andere, das sich hinter dem Sichtbaren verstecken kann, herauszulesen (Physiognomik), gibt es ja eine alte Erfahrung und eine fast unübersehbare Literatur. Dasselbe gilt gewissermaßen auch vom Auge, dessen wechselnder Ausdruck so viele Künstler und Photographen eingefangen haben (vgl. den Artikel von Dr. NOELLE CHOMEE in B. BELLUGUES Darstellung in "Formes et Couleurs" 19). Auch vom menschlichen Munde gilt dasselbe, obgleich vielleicht in geringerem Grade. Die Hand des Menschen, bisweilen so ausdrucksvoll, oft in noch höherem Grade als das Gesicht die unbewußte Visitenkarte des Menschen, hat seit ältesten Zeiten einen Symbolwert, der kaum übertroffen werden kann. Man gibt einander die Hand, um ein Versprechen zu bekräftigen. Die geballte Hand zeigt Festigkeit, die erhobene geballte Faust bedeutet Haß oder Drohung. Mit der Hand schreibt man seine Namensunterschrift. In meiner Kindheit sah ich bisweilen, wie einem nicht Schreibfähigen von jemandem geholfen wurde, der seinen Namen schrieb, und darunter die Bemerkung: "Mit der Hand auf der Feder". Darauf setzte der nicht Schreibkundige seine Hausmarke auf das wichtige Papier

    The study of RNA interference, editing, and modification has led to major shifts in our understanding of how genes are expressed and regulated. In RNA Interference, Editing, and Modification: Methods and Protocols, hands-on experimentalists describe in detail the protocols and assays they have developed to study these processes in most of the major biological systems in which they are known to occur-a wide range of organisms that includes worms, flies, trypanosomes, mammals, and plants. A historical overview of each subject is provided to put related chapters into a larger context. RNA interference (RNAi) has also emerged in recent years as an important and promising tool for gene discovery and reverse genetic engineering. Application of this cutting-edge technique is described for a range of experimental systems. Topics of interest include stable and transient RNA interference, RNA editing, gene silencing, bioinformatics, small noncoding RNAs, and RNomics. Special attention is given to methods for the identification and characterization of small RNAs, many of which are involved in RNA interference or modification. In addition, techniques used to study RNA editing mechanisms are elaborated for both systems that involve the conversion of one base to another and insertion-deletion editing. To promote the adaptation of their assays and approaches for use in other biological systems, the authors have included a range of methods representative of the major experimental systems in a given field. Poisons attract attention because they are both dangerous and  hard to discover. Secretive and invisible, they are a challenging object of representation. How do science studies, literature, and especially film-the medium of the visible-explain and show what is hidden? How can we deal with uncertainties emerging from the ambivalence of dangerous substances? These considerations lead the editors of this volume to the notion of “precarious identities” as a key discursive marker of poisons and related substances. This book is unique in facilitating a multi-faceted conversation between disciplines. It draws on examples from historical cases of poisoning; figurations of uncertainty and blurred boundaries in literature; and cinematic examples, from early cinema and arthouse to documentary and blockbuster.     Henri Grosjean Naturally occurring RNA always contains numerous biochemically altered nucleotides. They are formed by enzymatic modification of the primary transcripts during the complex RNA maturation process designated RNA modification. A large number of enzymes catalyzing the formation of these modified nucleosides or converting one canonical base into another at the posttranscriptional level have been studied for many years, but only recently have systematic and comparative studies begun. The functions of individual enzymes and/or the modified/edited nucleosides in RNA, however, have remained largely ignored.This book provides advance information on RNA modification, including the associated editing machinery, while offering the reader some perspective on the significance of such modifications in fine-tuning the structure and functions of mature RNA molecules and hence the ability to influence the efficiency and accuracy of genetic expression.
